This method of colouring a raster layer will not work on masks, backgrounds, adjustment layers or vectors. Highlight the layer you wish to work on in your layer pallet. Notice the grey lock in the right of your layer pallet.  Click on it to lock the transparency. The lock will turn blue.  You can now use the flood fill tool to colour your layer.  Click on the lock once more to unlock it. It will turn grey again.
